FAQs – Protein Quantification with SPEAR
The SPEAR UltraDetect™ assay kit contains qPCR positive control to ensure the reliability of protein quantification in addition to having a rigorously tested calibration curve. Assay specific supplementary kits, containing extra lyophilized calibrator and assay diluent, are available for purchase if interested in contriving unique control sets.
PCR Positive Control: This control comes in lyophilized form and can be reconstituted in buffer, just like a sample. It is used to validate the PCR amplification step during, ensuring that the assay is functioning correctly.
SPEAR UltraDetect™ assays don’t require any proprietary instrumentation. SPEAR utilizes standard qPCR for protein quantification readout. However, the full SPEAR UltraDetect™ immunoassay solution includes the F.A.S.T™ liquid handler from Formulatrix® and the BioShake iQ from QInstruments.
SPEAR protein quantification results are analyzed with SPEARview software to quickly and easily convert qPCR results into protein quantification results.
Our protein quantification workflow requires about 3.5 hours to complete, using sub-μL samples. This includes qPCR run time, and protein quantification results using SPEARview software, which converts qPCR cycle results into protein quantification.
Protein quantification with SPEAR UltraDetect™ utilizes F.A.S.T™ liquid handler from Formulatrix, which performs all reagent additions as well as sample and calibrator dilutions. Typical user hands-on time is under 30 minutes.
