Ultrasensitive α-Synuclein and pS129-α-Synuclein Assays for Biomarker Research

α-Synuclein aggregation and misfolding are central pathological features of synucleinopathies, including Parkinson’s disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA). But detecting disease-relevant α-synuclein species in biofluids remains challenging due to low endogenous abundance, complex sample matrices, and the need to measure subtle changes across disease progression.

The SPEAR UltraDetect™ α-syn and pS129-α-syn assays were developed to quantify total α-synuclein and phosphorylated α-synuclein at Ser129 with femtogram-level sensitivity, high precision, and broad matrix compatibility across plasma, serum, and CSF.

In this application note, you’ll learn how SPEAR UltraDetect™ α-synuclein assays deliver:

  • Femtogram-level detection with a 5-log dynamic range, enabling reliable quantification of low-abundance biomarkers, including EV-based applications.
  • High analytical precision with average CV < 5%, supporting confident detection of subtle biological changes and longitudinal biomarker monitoring.
  • Flexible analysis options, enabling individual biomarker quantification or pS129/total α-syn ratio-based analysis using singleplex or parallel plex workflows.
  • Minimal sample input, requiring just 1 μL of diluted sample per replicate to preserve precious plasma, serum, and CSF samples.
  • 100% quantifiability of endogenous samples from healthy individuals across plasma, serum, and CSF.
  • Differentiation of Parkinson’s disease from healthy controls using total α-syn, pS129-α-syn, or the composite pS129/total α-syn ratio.
